内容 抄録:In the living body, cells are often exposed to acid environment such as metabolic or respiratory acidosis, inflammation loci and insight of solid tumors. However the mechanisms of cell adaptation to extracellular acid environment have not been investigated until now. For the past several years, some exchangers such as Na^+/H^+ exchangers and HCO_3^--dependent alkalinizing transporters have been reported to perform important roles in intracellular pH recovery after acute acid load. Here I showed that Jurkat cells proliferated, with maintaining intracellular pH under extracellular acidic conditions and these transporters were not involved in the pH homeostasis. Ouabain and N-ethylmaleimide reduced pHi, so it is possible that ouabain or N-ethylmaleirnide sensitive proteins may have an important role in intracellular pH maintenance. Indeed, our data showed that there are N-ethylmaleimide sensitive K^+-ATPases and N-ethylmaleimide binding proteins those expressions were changed under low extracellular pH. Here I also showed that Jurkat cells change intracellular signal pathways under acidic pH conditions. Phosphotyrosine level of some proteins are changed during proliferation in acidic media, MAPK activity was also changed rapidly as lowering of extracellular pH but the change disappeared as time passed. These results suggest that Jurkat cells may be adapted to acidic environment with changing protein phosphotyrosine level, MAPK pathway and finally expression of some genes required for adaptation to extracellular acidic environments.
