内容 The plate culture method offers a means of isolating pure species of bacteria in a comparatively simple manner. In the case of spiral bacteria, this method is expected to be used in principle. As compared with the other contaminating bacteria (rods and cocci), the spiral bacteria grow so slowly on a solid medium that the colony becomes visible to the naked eye after the elapse of at least 5 days. In view of this fact, if the plate culture method is employed to isolate the spiral species in pure culture, many colonies deriving from the contaminating bacteria are formed previous to the colonies of the spiral bacteria. Consequently, the development of the colonies of spiral bacteria is checked and their pure culture tends to be impossible. Moreover, many investigators have found it very difficult to distinguish the colonies of spiral bacteria from those formed by the other bacteria. Hence, the difficulty of pure culture of the spiral bacteria prevents the study of it and few reports have been published. This paper presents a simple method of pure culture of halophilic spirillum (Spirillum) and purple spiral bacterium (Rhodospirillum).
