Enzymatic Synthesis of New Glycosides by Transgalactosylation with Escherichia coli β-Galactosidase   :   Escherichia coli由来β-galactosidaseの糖転移作用による新規配糖体の酵素的合成

内容 β-D-xylosyl glycerol, lactoseを含む溶液中でEscherichia coli β-galactosidaseを作用させ, その反応液を活性炭およびセルロースカラムクロマトグラフ法に供したところ, ο-β-D-galactosyl-(1→4)-ο-β-D-xylosyl-(1→1)-glycerol, ο-β-D-galactosyl-(1→4)-ο-β-D-xylosyl-(1→2)-glycerol, ο-β-D-galactosyl-(1→4)-ο-β-D-xylosyl(1→3)-glycerolの3種の異性体からなる新規配糖体標品が得られた.
The enzymatic synthesis of new glycosides from β-D-xylosyl glycerol (XG) and lactose by transgalactosylation with Escherichia coli β-galactosidase was studied. For the transgalactosyl reaction, a reaction mixture containing XG, lactose, and the β-galactosidase was incubated at 37℃ for 0.5 h. New glycosides were isolated by charcoal and cellulose column chromatography and identified as the mixture of three isomers; ο-β-D-galactosyl-(1→4)-ο-β-D-xylosyl-(1→1)-glycerol, ο-β-D-galactosyl-(1→4)-ο-β-D-xylosyl-(1→2)-glycerol and ο-β-D-galactosyl-(1→4)-ο-β-D-xylosyl-(1→3)-glycerol.
